ACE-031
ACE-031 (also known as ACVR2B-Fc) is an engineered fusion protein designed as a soluble decoy receptor that inhibits myostatin (GDF-8) and other related signaling proteins in the Transforming Growth Factor-beta (TGF-\beta) superfamily.
Originally developed by Acceleron Pharma and Shire to treat muscle-wasting diseases like Duchenne Muscular Dystrophy (DMD), ACE-031 is widely known in research circles as one of the most potent myostatin inhibitors ever produced.
How ACE-031 Works: The Decoy Mechanism
Myostatin is a naturally occurring protein that acts as the body’s "brakes" on muscle growth, preventing skeletal muscle from growing excessively. Under normal conditions, myostatin circulating in the blood binds to the Activin Type IIB Receptor (ActRIIB) on muscle cell membranes, signaling the cell to limit muscle synthesis.
ACE-031 alters this process through a trap mechanism:
1. Soluble Decoy Binding: ACE-031 consists of the extracellular binding portion of the ActRIIB receptor fused to a human antibody fragment (Fc region).
2. Ligand Trapping: When introduced into circulation, ACE-031 floats freely and binds to circulating myostatin, Activin A, Activin B, and GDF-11 before they can reach the actual receptors on muscle tissue.
3. Uninhibited Growth Signal: By trapping these growth-limiting proteins, ACE-031 effectively "cuts the brakes," allowing muscle satellite cells to proliferate and hypertrophy without native biological restrictions.
