Melanotan 2 (also known as MT-2) is a synthetic cyclic heptapeptide (Ac-Nle-cyclo[Asp-His-D-Phe-Arg-Trp-Lys]-NHâ‚‚).
Like Melanotan 1, it is an analogue of the naturally occurring peptide hormone \alpha-MSH (\alpha-melanocyte-stimulating hormone). However, its cyclic structure grants it a much higher potency, a longer biological half-life, and broad affinity across multiple central and peripheral melanocortin receptors.
Primary Key Mechanisms
 Broad Melanocortin Receptor Agonism: Unlike MT-1 (which primarily targets MC1R in the skin), MT-2 non-selectively stimulates MC1R, MC3R, MC4R, and MC5R.
 Melanogenesis (MC1R): Binds heavily to MC1R on epidermal melanocytes, triggering tyrosinase activity to convert pheomelanin into photoprotective eumelanin (dark brown pigment), producing rapid skin tanning with minimal UV exposure.
 Pro-Erectile & Sexual Function (MC3R/MC4R): Crosses the blood-brain barrier to bind central MC3R and MC4R in the hypothalamus, inducing central nervous system-mediated sexual arousal and erectile responses independent of vascular pathways.
 Appetite Suppression (MC4R): Central MC4R activation signals satiety in the brain, leading to a notable reduction in appetite and reduced caloric intake.
