PT-141 (brand name Vyleesi, generic name Bremelanotide) is a synthetic cyclic heptapeptide (Ac-Nle-cyclo[Asp-His-D-Phe-Arg-Trp-Lys]-OH).

Originally developed as a metabolite derivative of Melanotan 2 (MT-2), scientists isolated PT-141 specifically to harness its pro-sexual signaling while stripping away the N-terminal amide group, removing MT-2’s severe skin-tanning properties. It is a targeted therapy designed for the treatment of sexual dysfunction in both men and women.

Primary Key Mechanisms

Β Central Nervous System Activation: Unlike traditional vascular medications (like PDE5 inhibitors/Viagra) that act peripherally on blood vessels, PT-141 acts directly on the central nervous system by crossing the blood-brain barrier.

Β Melanocortin Receptor Agonism: It functions primarily as a potent agonist at MC3R and MC4R (melanocortin 3 and 4 receptors) in the hypothalamusβ€”the region of the brain regulating motivation, desire, and sexual behavior.

Β Dopaminergic Pathways: Binding to hypothalamic MC4R triggers downstream release of dopamine, directly heightening sexual desire, arousal, and intimacy-seeking behavior.

Β Minimization of Melanogenesis: By removing the structural components that drive heavy MC1R activation, PT-141 produces minimal to no skin darkening compared to its parent compound, MT-2.
